Most of us are familiar with Phase I, II, III, and IV clinical trials. But before these phases, there is another stage called Phase 0, also known as a human microdosing study. It is one of the least discussed phases, yet it plays an important role in modern drug development.
In a Phase 0 trial, a small group of healthy volunteers receives a very tiny dose of an investigational drug. The dose is much lower than the amount needed to treat a disease, so the medicine is not expected to produce any therapeutic effect.
The main purpose is simple: researchers want to understand how the drug behaves inside the human body. They study whether the drug is absorbed, how it travels through the body, how it is broken down, and how it is eliminated. This helps them determine whether the results seen in laboratory and animal studies are likely to translate to humans.
One thing I found interesting is that Phase 0 is not about proving that a drug works. Instead, it helps researchers decide whether the drug is promising enough to move on to Phase I clinical trials. If the drug doesn’t behave as expected, its development can be stopped early, saving valuable time, money, and research efforts.
Although Phase 0 provides useful information, it cannot answer questions about the drug’s safety, effectiveness, or the right dose. Those are evaluated in the later phases of clinical trials.
I think Phase 0 is a great example of how even a small study can make a big impact. By gathering early human data, researchers can make smarter decisions and improve the efficiency of the drug development process.
