Recently, IIT Bombay has developed and demonstrated a gentler version for recovering lab-grown T cells from 3D scaffolds. This is particularly relevant to CAR (Chimeric Antigen Receptor) T-cell therapy for treating cancer, but is also useful in other immunotherapy and tissue engineering research. Traditionally, Simple Manual Flushing and Harsh Enzymatic Digestion were the methods used for this purpose.
What is CAR T-cell Therapy?
This therapy treats blood cancers by using the patient’s own immune cells, specifically T-cells, and modifying them to recognize and attack cancer cells.
Step-by-Step Process:
- Leukapheresis: Collecting T-cells from the patient’s blood using an apheresis machine and sending them to a laboratory
- Lymphodepleting Chemotherapy: To reduce the patient’s existing lymphocytes in their body to create an optimal environment for the modified T-cells to multiply
- CAR T-cell Manufacturing: Chimeric Antigen Receptor T-cells are manufactured
- Infusion: CAR T-cells are administered into the patient’s bloodstream
Why is Accutase Better than Traditional Methods in Recovering T-cells from Scaffolds?
- Accutase works as a gentler enzymatic dissociation
- Keeping the CAR T-cells intact without destroying them
- Higher cell recovery
- Better cell sustainability
