In 2025, a six-year-old girl in China became the world’s first person to receive a **brain-targeted CRISPR base-editing therapy** for a rare genetic condition. It was a moment that reflected promise and potential gene-modulation technology.

She had **Snijders-Blok-Campeau syndrome** , a rare condition caused by mutations in the **CHD3 gene**. While the disorder can cause developmental delay and intellectual disability, it is not usually life-threatening.

### **What did the treatment involve?**

The therapy was developed by neuroscientist Zilong Qiu and his team at Xinhua Hospital.

It used **CRISPR** base editing, delivered to the brain through **adeno-associated virus (AAV) vectors** injected into the cerebrospinal fluid.

The child’s parents reportedly contributed around US$860,000 to help fund the personalized treatment.

### **What Happened?**

Just within 7 days of recieving the treatment, she developed severe complications, including a high fever, kidney injury, and an intense inflammatory response. She sadly passed away a week later. An ongoing investigation suggests that the immune system’s reaction to the viral delivery system may have played a role.

### **Concerns Raised**

The case has sparked difficult but important questions. Investigators allege that the patient’s death was not publicly reported, trial records were not updated, and subsequent scientific publications did not mention the fatal outcome or certain financial disclosures. These allegations have fueled discussions about transparency in medical research.

### **Why Does This Matter?**

Every breakthrough in medicine carries responsibility. Cases like this remind us why informed consent, honest reporting of adverse events, and rigorous ethical oversight are just as important as scientific innovation itself. Public trust in research depends on these principles.

### **Does This Mean Gene Therapy Is Unsafe?**

**Not at all.**

Gene therapy has already changed the lives of many people living with rare genetic disorders. Rather than questioning the technology itself, this case highlights the need for careful safety evaluation, transparent communication, and strong ethical standards—especially when pioneering treatments are being tested in humans.

**Scientific progress is built on both successes and setbacks. The true measure of progress lies not only in celebrating breakthroughs but also in learning openly from the challenges along the way.**

**What are your thoughts on this?**

Scientific innovation should always go hand in hand with transparency and ethics. **Every setback provides valuable lessons but those lessons can only benefit future patients if adverse outcomes are reported openly and honestly. Trust is just as important as technology.**

Trust, transparency and ethics work hand-in-hand. This incident emphasized the importance of Informed consent. In this case, parents are particularly concerned about the doctors’ perceived ignorance in publicly disclosing results.

Despite the lack of success in the therapy, the case study gives us significant insight into the difficulties of this methodology. Herein lies the importance of the failure as a learning experience.

There are many instances in which gene therapy can prove to be successful. However, delivering gene therapy into the brain using CSF is complicated. The body’s immune system can identify the viral delivery system as foreign and mount an immune response that activates certain immune cells. This can decrease the efficiency of the delivery system. it may also cause inflammation or other immune-related effects.

Failure is a learning experience but this failure took a life, that is the most heart-breaking aspect of this incident.

And yes, injecting viral vectors directly into the cerebrospinal fluid is indeed complicated and may evoke a great immune reaction.

A clinical trial is a very sensitive area of developing something new. So ethics and transparency are the pillars of the process. Sticking to the protocol and taking no shortcuts are good. I don’t know what exactly happened in this case, but whatever the outcome is, it should be shared to prevent another such case.

There’s a muted response and the university where the trial was conducted paid a fine of more than a half-million dollars and had further gene therapies suspended.

But the child’s parents still mourn for their loss.

I agree. Every failed experiment can provide valuable information. Progress happens when we remain curious, learn from what didn’t work, and keep improving.

Always, the victim’s family suffers. In this case, it’s a little girl. That’s very sad.

This experiment didn’t work out, but it gave us important information. It also reminded everyone that the safety of people involved in research should always be the most important thing.
