Zombie cells (senescent cells), a contradictory topic, have a _ **heterogeneous nature** _ (“double-edged sword”); they can _prevent cancer_ for years while also _ **accelerate** _ the formation of malignant cells. That creates the most sensitive space for research. Some studies also highlighted the role of ZC in age-related disorders such as _ **Alzheimer’s** _, _ **dementia** _, _osteoporosis_, etc., and persistent inflammation linked to the aging process. Their buildup also prompts the emergence of a variety of chronic illness. > Normal cells either die or live in cellular apoptosis, whereas “_ **zombie cells** _” remain metabolically active but no longer divide. The motive of senescence is to impede the division of damaged cells; conversely, during this period, they discreetly adapt to more _ **aggressive** _ cell division. _Targeting zombie_ cells could be a promising cancer treatment approach. ![cellular senescence](https://canada1.discourse-cdn.com/flex033/uploads/medbound/original/2X/6/6c1c6298775f2eb22d0fec00ac12bb350060f02b.jpeg) **Mechanism** ZC possesses a proinflammatory secret senescence-associated secretory phenotype (SASP) and enters a long-lasting growth arrest gateway, releasing **cytokines** , **chemokines** , and **matrix metalloproteinases** while also triggering epigenetic alterations in **histones** and **DNA**. **Tumorigenic impact** ▪Creating new blood vessels—MMP-2, MMP-9, and VEGR-A. ▪Amplify local inflammation—Interleukin-6 and 8 suppress pro-apoptosis factors (BAX, PUMA, etc.) while activating Cyclin D1 and c-Myc, hence limiting tumorigenesis. ▪Metastasis—IL-6-mediated epithelial-mesenchymal transition (EMT) directs cancer cells to break away from parent tumors and move via the bloodstream to form new tumors in distinct organs. **Anti-tumorigenic impact** 🔹Immune Activation: To eradicate senescence and premalignant cells, which are crucial to the initial stage of neoplastic cells, SASP first attracts natural killer cells and other macrophages. 🔹Tumor suppression: Damaged DNA plays a key role in stopping the proliferation of the cells. I go through a research study that claims zombies have a _ **protective protein barrier** _ known as “ **GPX4** ” that may even resist fatal _ **ferroptosis** _, a kind of cell death linked to elevated iron levels and the destructive reactive oxygen species. Ferroptosis poses a serious _ **threat** _ to senescence cells. By blocking the GPX4 protective shelf, zombie cells can die. Researchers have tested the drug in a mouse and observed that eliminate senescence \*halts tumorigeneses and boost survival rate.
